Bioreactor cultivation of CHO DP-12 cells under sodium butyrate treatment – comparative transcriptome analysis with CHO cDNA microarrays
نویسندگان
چکیده
Background Sodium butyrate (NaBu) is not only known to inhibit proliferation but also to increase the specific productivity in cultivation of Chinese hamster ovary (CHO) cells [1] – the most commonly used mammalian cell line for pharmaceutical protein production [2]. So far, little is known about the underlying mechanisms and genes that are affected by butyrate treatment. Besides the proteomic approach to unravel proteins involved in the processes, the analysis of transcriptomes presents another promising method. Here we show an application of our CHO cDNA microarray to identify genes associated with increased productivity during cultivation of CHO cells under sodium butyrate treatment.

Next-generation sequencing of the CHO cell transcriptome
Since 1957 Chinese hamster ovary (CHO) cells are used for in vitro cultivation as they require assimilable low sustenance [1]. Today, CHO cell lines represent the most commonly used mammalian expression system for the production of therapeutic proteins and are considered as the mammalian equivalent of E. coli in research and biotechnology [2]. The production of biopharmaceuticals in CHO cells i...

Background: IFN-g is mostly secreted by activated CD4+ , CD8+ T cells and NK cells. This cytokine has immunomodulatory, anti-cancer and anti-microbial effects and is important for prophylaxis, diagnosis and treatment of chronic infections and cancers. Objective: The purpose of this study was to clone the full cDNA of human IFN-g and express it in CHO cell line. Methods: Lymphocytes from a healt...

Background For industrial pharmaceutical protein production fast growing, high producing and robust cell lines are required. To select more pH shift permissive and fast growing sub-populations, the CHO DP-12 (ATCC clone #1934) cell line, an anti-IL8 antibody producing CHO K1 (DHFR) clone, was continuously subcultured at high viability (>90 %) for more than four hundred days in shaking flasks us...
متن کاملModulation of heparan sulfate biosynthesis by sodium butyrate in recombinant CHO cells.
Sodium butyrate, a histone deacetylase inhibitor, has been used to improve transgene expression in Chinese hamster ovary (CHO) cells. The current study explores the impact of butyrate treatment on heparan sulfate (HS) biosynthesis and structural composition in a recombinant CHO-S cell line expressing enzymes in the heparin (HP)/(HS) biosynthetic pathway (Dual-10 stably expressing NDST2 and HS3s...
متن کاملA kinetic-metabolic model based on cell energetic state: study of CHO cell behavior under Na-butyrate stimulation.
A kinetic-metabolic model approach describing and simulating Chinese hamster ovary (CHO) cell behavior is presented. The model includes glycolysis, pentose phosphate pathway, TCA cycle, respiratory chain, redox state and energetic metabolism. Growth kinetic is defined as a function of the major precursors for the synthesis of cell building blocks. Michaelis-Menten type kinetic is used for metab...
